A PCR-based survey of animal African trypanosomosis and selected piroplasm parasites of cattle and goats in Zambia OAK

We screened cattle and goats from the districts of Chama, Monze and Mumbwa in Zambia for animal African trypanosomes, Babesia bigemina and Theileria parva using PCRs; 38.1% of the samples tested positive for at least one of the parasite species. The most common parasite was Trypanosoma vivax (19.8%). Its incidence was significantly higher in goats than in cattle, (P<0.05). B. bigemina was found in samples from all the three areas, making it the most widespread of the parasites in Zambia. Among the tested samples, 12.0% of the positive samples were mixed infections. A trypanosome species isolated from naturally infected Haemaphysalis hystricis ticks in Kagoshima Prefecture, Japan. OAK

Common arthropod vectors for trypanosomes are flies, fleas and bugs. This study reports on an unknown trypanosome species isolated from naturally infected Haemaphysalis hystricis ticks, hereby, referred to as Trypanosoma KG1 isolate. The parasite has been successfully cultured in vitro with L929 or HEK 293T cell line as feeder cells. This trypanosome cannot survive in vitro without feeder cells. Following experimental infections of ticks, the trypomastigote-like and the epimastigote-like forms of this trypanosome could be detected by Giemsa-stained smears of the midgut and salivary glands of Ornithodoros moubata ticks which were made to feed on a culturing medium containing Trypanosoma KG1 isolate through an artificial membrane. Advances in the artificial feeding of Ornithodoros moubata (Acari: Ixodidae) and the follow up of its life cycle after feeding on fetal bovine serum OAK

Ornithodoros moubata was fed artificially through a parafilm membrane with two types of artificial meals. The first type was composed of bovine and horse red blood cells suspended in phosphate buffered saline or fetal bovine serum (FBS). The second type was composed of FBS alone. Engorgement and post engorgement survival rates of the ticks after feeding on the two types of meals were compared and analysed statistically. FBS was found to be superior to red blood cell meals. Eventually two batches of FBS from different manufacturers were used to feed the ticks through artificial membrane. Characterization of an epimastigote-stage-specific hemoglobin receptor of Trypanosoma congolense OAK

Background: Since Trypanosorna spp. lack a complete heme synthesis pathway, the parasites are totally dependent on their host for heme throughout all of the stages of their life -cycle. We herein report the identification and characterization of a T. congolense epimastigote form (EMF)-specific hemoglobin (Hb) receptor. The gene was initially reported to encode a T. congolense haptoglobin (Hp)-Hb complex receptor (TcHpHbR) based on its similarity to a gene encoding a T brucei Hp-Hb complex receptor (TbHpHbR). Prompt and accurate diagnosis of malarial patients is a crucial factor in controlling the morbidity and mortality of the disease. Effective treatment decisions require a correct diagnosis among mixed-species malarial patients. Differential diagnosis is particularly important in cases of Plasmodium vivax, a species that shares endemicity with P. falciparum in most endemic areas. Moreover, it is difficult to identify P. knowlesi on the basis of morphology alone, and rapid diagnostic tests are still not available for this malaria species. Diagnosis of Babesia caballi Infection in Horses by Polymerase Chain Reaction OAK

A set of primers were designed according to the published sequence of the gene encoding a rhoptry protein of Babesia caballi, and used to amplify parasite DNA from the blood samples obtained from carrier horses by polymerase chain reaction(PCR)method.The PCR method was sensitive enough to detect parasite DNA from 2.5 μl blood sample with a parasitemia of 0.000001%. The PCR method was compared with fluorescent antibody test(IFAT) in order to evaluate the diagnosis effciency for B. caball infection in horses. Of 142 field samples from Mongolia, 28(20%) and 96(69%)samples were identified positively by PCR and IFAT, respectively. Exposure to 50-Hz electric fields on stress response initiated by infection with the protozoan parasite, Toxoplasma gondii, in mice OAK

The present study was aimed to examine whether a 50-Hz electric field (EF) affects biological stress responses. Male C57BL/6J mice were exposed to either 50-Hz EF (45 kV/m) or given a sham exposure for 30 minutes per day for 1, 2 or 4 weeks and then either mock inoculated or inoculated with the protozoan parasite, Toxoplasma gondii (T. gondii) PLK strain. Survival rate was calculated in mice infected with T. gondii after exposure to EF and the serum antibody levels to heat shock protein 60 (Hsp-60), and mRNA levels for Hsp-60, IFN-γ and IL-12p35 in mouse peripheral leucocytes were determined. Loop-mediated isothermal amplification (LAMP) assays for detection of Theileria parva infections targeting the PIM and p150 genes OAK

We have developed two loop-mediated isothermal amplification (LAMP) assays for the detection of Theileria parva, the causative agent of East Coast fever (ECF), an economically important cattle disease in eastern, central and southern Africa. These assays target the polymorphic immunodominant molecule (PIM) and p150 LAMP genes. The primer set for each gene target consists of six primers, and each set recognises eight distinct regions on the target gene to give highly specific detection of T. parva. The detection limit of each primer set is 1 fg, which is equivalent to one copy of the PIM and p150 T. parva genes. Mycophenolic acid and its derivatives as potential chemotherapeutic agents targeting inosine monophosphate dehydrogenase in Trypanosoma congolense OAK

This study aimed to evaluate the trypanocidal activity of mycophenolic acid (MPA) and its derivatives for Trypanosoma congolense. The proliferation of T. congolense was completely inhibited by adding <1 mu M MPA and its derivatives. In addition, the IMP dehydrogenase in T. congolense was molecularly characterized as the target of these compounds. The results suggest that MPA and its derivatives have the potential to be new candidates as novel trypanocidal drugs.
